I am trying to get an old Mettler (DL38) up and running and am having trouble getting it to blank (pre-titrate). Once it does blank and states "ready" it drifts back so quickly that I am unable to even determine an adequate concentration for my Comp5 or get an accurate standard reading. I am purifying a six-His tagged 50kDa hnRNP K protein from E coli cells using IMAC with Cobalt TALON resin. I am trying to send the proteins for crystallography and it requires a good quality of sample. My problem is that smaller bands are visible on the gel and I don't know how to get rid of them.
What might the problem and how can I fix it?

SSE exceed 1GW renewable energy generating capacity….

Good news for SSE this month as they have hit a huge milestone in their production of renewable energy. SSE has managed to build enough onshore wind farms throughout the UK to generate power for 700,000 homes.
It is believed that the generating capacity of the SSE wind farms has now exceeded 1GW.
This news comes even as SSE are looking to increase generating capacity even further by building onshore farms in Northern Ireland, Ireland and Scotland.
The Chief Executive of SSE Ian Marchant has made it clear he wants to continue to grow capacity “significantly” in the coming months.In the past 6 years SSE’s onshore generating capacity has increased massively from 40MW to exceed their more conventional hydro electric capacity, of 1,150MW.
And with construction work ongoing in Northern Ireland, Ireland and Scotland it is expected that the generating capacity of SSE is only going to increase in coming months.
The real question is what are the rest of the big six energy companies doing to follow this example?
SSE is clearly betting big on the renewable future of the UK, do they know something the others don’t? Or are the others just working away quietly whilst SSE courts all the publicity?

Hi everybody, I have problem to detect 2KDa oligopetide on SDS PAGE. I made 17% Tris-Glycine gel and stained with CBB but didn't work as usual. Anybody does have a protocol for low MW peptide? I' going to do western blot on this SDS PAGE. I heard about Tricine gel but don't have all stuff and no money to buy them. Thanks in advance -
weird sine-wave oscillations?

Hello,
I was recording some ligand-gated ionic currents with a voltage step protocol, and I got puzzled, why are all these sine wave-like oscillations appearing at the negative potentials that seemingly do not carry any channel currents? Are they normal, or are they just instrumental noise? coz I think I definitely have some channel-mediated current at some more positive potentials on the left one.
Just wonder if I can get any help?

Question on Schaffer Collateral

Hi,
I'm new to electrophysiology techniques as I did my undergrad in Human genetics. Right now I'm trining in 'in vivo' electrophysiology techniques. Lowering a bipolar stimulation electrode and a recording electrode into the hippocampus to induce LTP by using three trains of HFS at 200 hrz within 5 minute intervals at the Schaffer Collateral.

Freezing and Thawing cancer cells

I have been freezing cells in a freezing medium consisting 10% DMSO, 20% FBS and 70% RPMI or DMEM. The cells will be stored overnight at -80oC and then transfered to liquid N2 . We make our own freezing medium as indicated above. The problem comes when we want to use these cells after storage. All the cells are dead and there are no viable ones despite storage at the appropriate density and having been stored at greater than 90% viability. W... -

Problem in repeatability of Real time PCR results

Dear Friends,
We are working on HBV quantification by using Rotor gene with Artus HBV PCR kit, the problem here is we are trying to work in 1/4 of the recommended volume and it is not showing similar results with the full volume results.
We have taken 7.5 microliters of master mix and 5 microliter of Extracted DNA and the recommended volume is 30 microliter of master mix and 20 microliter of extracted DNA

High Hg levels in blood

I am using Agilent 7700x (He collision mode) for Hg quantification in blood.
No memory effects we have.Calibration curve is linear,recovery is correct, QC and background are good.
But when we read "normal" samples (expected concentration below 5 ppb) ,in some samples , (20-30%) ,we found values about 10-15 ppb. Wht?s hapenning? .

I'm new to the whole virus studying world and I'm working on a line of cells that are new to me, MDCK cells. Is it true that if they become over confluent they start to transform?
I've been having a hard time finding images of transformed vs non-transformed MDCK cells.

I am hoping to start culturing a new human prostate cell line (MDA-PCa-2B) that is a bit troublesome in culture. ATCC suggests making an alternative media that contains a laundry list of items including bovine insulin. I am currently having a hard time getting bovine insulin from our local supplies and was wondeirng if anyone knows of a good substitution. Can i substitute insulin from another species? -

Hallo! I need to purify a very hydrophobic, transmembrane protein (30-40 kDA). It is actually one in a family of related proteins. The goal would be to be able to separate all of them, but I would start with one.
In the literature I found people separating one of these proteins with PRP-3 from Hamilton (300 A I think ). In the Hamilton web site I found PRP-3 columns 10 or 12-20 uM. Which one would be more suitable to my study? WEhant about the column lenght?

Transduction troubleshooting: low efficiency

I am currently trying to get good transfection efficiency after infection of the bone marrow cells with the ecotropic murine stem cell retrovirus (MSCV). In brief, I have transfected my ecotropic virus (MSCV) into 293T cells and tested for GFP expression after growing then in NIH3T3 cells. Luckily, I got 90-95 % positive for GFP, analyzed by FACS. However, when the same virus is transduced into primary bone marrow cells by spinfection at 2000 rpm for 1.5 h, I am only getting ... -

Be 6.8
Co 8.5
In 8.5
and still pass the SmartTune Autolens? I was assuming that the DAC values had to go up linearly. Is this something that would affect Mn not passing a calibration curve????

I read a fantastic article on the Business Green Industry Voice Blog today about how the London Olympics has the potential to make some permanent changes regarding Londoners approach to a more sustainable way of life. As it says in the blog post “It is easier to get people to think about changing their behaviour when their normal routines are disrupted.”
The main disruptions look like they will form around transport givenm that there is to be an expected influx of around 5.3 million visitors over the 100 day affair. In fact on some of the more heavily scheduled days this will create a staggering 855,000 fan trips. So with over 100 miles of Londons roads being shut off as the official Olympic Network things could get interesting.
