I'm wondering how to properly store bacterial cell lysates? Do you add protease inhibitors before putting them at -20C or lower temperatures? When will the samples last?
Because, I've done Western blots of the cell lysates. The first time I did it, the bands looked neat and I still could quantify the bands. But when I tried to do the blots for the 3rd and 4th time, I only saw smears.. What happened???

I am trying to make up fresh picrosirius red solution as the solutions currently in the lab are rather old.
The protocol I followed says add 500mg sirius red F3B to 500ml saturated aqueous picric acid. I bought in the saturated picric acid solution and added the dye, however it is not going into solution (presumably because it is already a saturated solution). Apparently this hasn't been a problem before.
Does anyone have any suggestions please?

I have isolated the Asipose Derived Stem Cells and differentiated them to the osteogenic differentiation with induction medium.The point is that I saw the lift off of the cells from the plate surface.
Is there any suggestion what would be the possible reason?

Hi all,
I am trying to intrduce my gene of interset by retroviral infection into MEFs. for this first I do transfection in platE cells and then collect the viral supernatant to infect MEFs. then i start the selection of infected MEFs. but the problem is, all my cells (MEFs) die during selection!!!!! I have tried everything till now, but nothing is working...

Hi ,
Please I need help!! I start working with a nested PCR, my samples are bronchoalveolar lavage and after DNA extraction I got A260/280 very low - around 1.41, 1.56 and sometimes A260/280 very high - around 7. I know a good ratio of ~ 1.8 is pure DNA. How can I interpret these results? Very low A260/280 seems that I don't have DNA? And higher results? I didn't get any band in my PCR with this samples.

Hi, I've been trying to culture primary hepatocytes but recovery after Percoll centrifugation is very low. In short, perfuse ~35 ml of Invitrogen Liver Digestion Media at 42°C. After braking apart, cells were suspend in plating medium and remove debris by passing a filter. After centrifugation, cells were suspended in 10 ml of plating medium. I'm having trouble identifying suspected gram + rods with API strips. I use 50 CH strips with CHB/E medium but sometimes run into a bacteria that I can't identify. I then put it on 20E strips but that doesn't give me results either. Are there other strips I can use beyond 50CH and 20 E strips if something looks like a gram + rod? Also what is the difference between CHB and CHE because the medium is specified as BOTH on the ampoule but then there is a profi...

We just recently started running a new Agilent 7700x ICP-MS and are having problems with bubbles in the line. This problem is affecting our RSD's. Has anyone else had this problem? We have tried different tubing and it has not helped. Any suggestions would be greatly appreciated.

I'm trying to align an 800-amino-acid sequence coded by a gene from a
known genome to a large set of ordered contig sequences from a new
genome of a related strain whose sequence is not yet fully resolved.
Using tblastn, I've so far been unsuccessful in finding

hi all,
I have done PCR using a universal primer and eluted the PCR product from the agarose gel using Nucleospin kit. Eluted pcr product on agarose gels is showing multiple bands on gel electrophoresis.
the PCR product of mine only showed single band but after elution am getting multiple bands...why am getting this multiple bands after elution?
