Hi, i will study nthy-ori 3-1 cell line (ECCAC number: 90011609) for my experiment. The problem is that i can't decide which kind of HBSS culture medium i should use. There are four main types which are:

I have amplify a gene with the size of 4.6 kb to 3 fragments and cloned them into Topo TA vector. All the 3 inserts have sticky ends when cut with restriction enzymes. I manage to cut the inserts from the respective vectors but I have a problem to ligate them. For your information, fragment a with the size of 1.2kb has a Sac II and ECoR1 site, fragment b with the size of 2.1kb has a ECoR1 and Pst 1 site and fragment c with the size of 1.5kb has a Pst1 and Not 1...

Hi,
i am using the double side gold seal . My instrument is 6890NB(FID). column is DB1, I use tempearature programme. The injector is 350, dectector final temerature is 360. I ananlyse hydrocarbon.
Cause using the double side gold seal, the nut is different from normal. but the tailing peking from solvent is more bigger than before.

I am working with Perkin Elemer ICP MS(Model 6100).Now I am facing a problem in plasma ignition.

I'm working in a molecular biology lab, doing multiple PCRs every day, and I am having a major problem with contamination in my water controls. (always the same size as the bands I'm looking for)
I've been trying to figure out why - new water, new primers, new mastermix, changing my gel loading technique, ect..., and I'll get rid of it but then it comes back. I can't think of what I'm doing to keep on bringing back contamination - anyone got sugg...

I am currently using a 5300DV ICP-OES from Perkin Elmer. I perform an acid digestion (nitric acid and hydrochloric acid) by means of open vessels. After digestion we dilute our samples to 50ml and filter them. I run the samples and identify the concentrations of 10 elements (Ca, K, Mg, Cu, Fe, Mn, Zn, S, Na and P). I am growing COS7 cells for an experiment and have the cells in T-25 flasks and they have been growing nicely. When I look at them under the microscope I see lots of cells, covering nearly the entire surface of the flask. Most of you will know that gold and silver are notoriously sticky causing memory effects. The resean is probably that the dissolved forms of these elements easily get reduced to elemental metals that ecipitate in teflon and tygon tubing.

Hi, i am trying to work out my kcat value and have the follwing information:
the enzyme concentration used was 0.007 mg per ml
the volume of enzyme added was 100ul into 3ml
the molecular weight of the enzyme is 149.5 kDa and it has 4 subunits
The vmax is 3.63 umoles min-1 ml-1
I know that kcat = vmax/[e]t but i am struggling to use the correct units as my kcat value needs to be in s-1

Hello,
I am trying to get an old Mettler (DL38) up and running and am having trouble getting it to blank (pre-titrate). Once it does blank and states "ready" it drifts back so quickly that I am unable to even determine an adequate concentration for my Comp5 or get an accurate standard reading. Anyone out there still using a DL38? Or can anyone help me with this issue. I know I've got vacuum on the sysytem because I don't have any trouble filling or emptying.......
